Detection of mecA gene and methicillin-resistant Staphylococcus aureus (MRSA) isolated from milk and risk factors from farms in Probolinggo, Indonesia

Background: Staphylococcus aureus is commonly found in dairy cows and a source of contamination milk. S. aureus that are resistant to beta-lactam antibiotics (especially cefoxitin) referred as methicillin-resistant Staphylococcus (MRSA). The spread MRSA cannot be separated from sanitation management during milking; it can originate milk collected the udder or hands farmers milking process. purpose this study was examine level cow's farmer's hand. Methods: A total 109 samples cow’s 41 farmer’s hand swabs were at farm Probolinggo, East Java, Indonesia. Samples cultured purified using mannitol salt agar (MSA). profile resistance established by disk diffusion test antibiotics, namely oxacillin cefoxitin. Results: The isolates that cefoxitin then tested for screening base (ORSAB) confirmation identity. suspected genotypically polymerase chain reaction (PCR) method detect presence mecA gene. results isolation identification 80 (53.33%) aureus. 42 (15%) 10 (12.5%) cefoxitin. ORSAB many 20 (47.62%) positive MRSA. In PCR testing gene, three (30%) gene. Conclusions: This shows several had gene encoding farms.
